X‐chromosome inactivation ensures dosage compensation between the sexes in mammals by randomly choosing one out of the two X chromosomes in females for inactivation. This process imposes a plethora of questions: How do cells count their X chromosome number and ensure that exactly one stays active? How do they randomly choose one of two identical X chromosomes for inactivation? And how do they stably maintain this state of monoallelic expression? Here, different regulatory concepts and their plausibility are evaluated in the (...) context of theoretical studies that have investigated threshold behavior, ultrasensitivity, and bistability through mathematical modeling. It is discussed how a twofold difference between a single and a double dose of X‐linked genes might be converted to an all‐or‐nothing response and how mutually exclusive expression can be initiated and maintained. Finally, candidate factors that might mediate the proposed regulatory principles are reviewed. (shrink)

BackgroundIn the Canadian Alliance for Healthy Hearts and Minds (CAHHM) cohort, participants underwent magnetic resonance imaging (MRI) of the brain, heart, and abdomen, that generated incidental findings (IFs). The approach to managing these unexpected results remain a complex issue. Our objectives were to describe the CAHHM policy for the management of IFs, to understand the impact of disclosing IFs to healthy research participants, and to reflect on the ethical obligations of researchers in future MRI studies.MethodsBetween 2013 and 2019, 8252 participants (...) (mean age 58 ± 9 years, 54% women) were recruited with a follow-up questionnaire administered to 909 participants (40% response rate) at 1-year. The CAHHM policy followed a restricted approach, whereby routine feedback on IFs was not provided. Only IFs of severe structural abnormalities were reported.ResultsSevere structural abnormalities occurred in 8.3% (95% confidence interval 7.7–8.9%) of participants, with the highest proportions found in the brain (4.2%) and abdomen (3.1%). The majority of participants (97%) informed of an IF reported no change in quality of life, with 3% of participants reporting that the knowledge of an IF negatively impacted their quality of life. Furthermore, 50% reported increased stress in learning about an IF, and in 95%, the discovery of an IF did not adversely impact his/her life insurance policy. Most participants (90%) would enrol in the study again and perceived the MRI scan to be beneficial, regardless of whether they were informed of IFs. While the implications of a restricted approach to IF management was perceived to be mostly positive, a degree of diagnostic misconception was present amongst participants, indicating the importance of a more thorough consent process to support participant autonomy.ConclusionThe management of IFs from research MRI scans remain a challenging issue, as participants may experience stress and a reduced quality of life when IFs are disclosed. The restricted approach to IF management in CAHHM demonstrated a fair fulfillment of the overarching ethical principles of respect for autonomy, concern for wellbeing, and justice. Cimpian et al. observed that we accept generic statements of the form ‘Gs are f’ on relatively weak evidence, but that if we are unfamiliar with group G and we learn a generic statement about it, we still treat it inferentially in a much stronger way: all Gs are f. This paper makes use of notions like ‘representativeness’, ‘contingency’ and ‘relative difference’ from psychology to provide a uniform semantics of generics that explains why people accept generics based on weak evidence. (...) The spirit of the approach has much in common with Leslie’s cognition-based ideas about generics, but the semantics will be grounded on a strengthening of Cohen’s relative readings of generic sentences. In contrast to Leslie and Cohen, we propose a uniform semantic analysis of generics. The basic intuition is that a generic of the form ‘Gs are f’ is true because f is typical for G, which means that f is valuably associated with G. We will make use of Kahneman and Tversky’s Heuristics and Biases approach, according to which people tend to confuse questions about probability with questions about representativeness, to explain pragmatically why people treat many generic statements inferentially in a much stronger way. Many philosophers appeal to intuitions to support some philosophical views. However, there is reason to be concerned about this practice as scientific evidence has documented systematic bias in philosophically relevant intuitions as a function of seemingly irrelevant features (e.g., personality). One popular defense used to insulate philosophers from these concerns holds that philosophical expertise eliminates the influence of these extraneous factors. Here, we test this assumption. We present data suggesting that verifiable philosophical expertise in the free will debate-as measured by (...) a reliable and validated test of expert knowledge-does not eliminate the influence of one important extraneous feature (i.e., the heritable personality trait extraversion) on judgments concerning freedom and moral responsibility. These results suggest that, in at least some important cases, the expertise defense fails. Implications for the practice of philosophy, experimental philosophy, and applied ethics are discussed. BACKGROUND -/- In biomedical ontologies, mereological relations have always been subject to special interest due to their high relevance in structural descriptions of anatomical entities, cells, and biomolecules. This paper investigates two important subrelations of has_proper_part, viz. the relation has_grain, which relates a collective entity to its multiply occurring uniform parts (e.g., water molecules in a portion of water), and the relation has_component, which relates a compound to its constituents (e.g., molecules to the atoms they consist of). -/- METHOD -/- (...) We distinguish between four kinds of complex entities and characterize them in first order logic. We then discuss whether similar characterizations could be given in description logics, and finally apply the results to mixtures. -/- RESULTS -/- At first sight, collectives and compounds seem to be disjoint categories. Their disjointness, however, relies on agreement about what are uniform entities, and thus on the granularity of description. For instance, the distinction between isomeric subtypes of a molecule can be important in one use case but might be neglected in another one. We demonstrate that, as implemented in the BioTop domain upper level ontology, equivalence or subsumption between different descriptions of same or similar entities cannot be achieved. Using OWL-DL, we propose a new design pattern that avoids primitive subrelations at the expense of more complex descriptions and thus supports the needed inferences. (shrink)

Background: SNOMED CT is a large terminology system designed to represent all aspects of healthcare. Its current form and content result from decades of bottom-up evolution. Due to SNOMED CT’s formal descriptions, it can be considered an ontology. The Basic Formal Ontology (BFO) is a foundational ontology that proposes a small set of disjoint, hierarchically ordered classes, supported by relations and axioms. In contrast, as a typical top-down endeavor, BFO was designed as a foundational framework for domain ontologies in the (...) natural sciences and related disciplines. Whereas it is mostly assumed that domain ontologies should be created as extensions of foundational ontologies, a post-hoc harmonization of consolidated domain ontologies in use, such as SNOMED CT, is known to be challenging. Methods: We explored the feasibility of harmonizing SNOMED CT with BFO, with a focus on the SNOMED CT Clinical Finding hierarchy. With more than 100,000 classes, it accounts for about one third of SNOMED CT’s content. In particular, we represented typical SNOMED CT finding/disorder concepts using description logics under BFO. Three representational patterns were created and the logical entailments analyzed. Results: Under a first scrutiny, the clinical intuition that diseases, disorders, signs and symptoms form a homogeneous ontological upper-level class appeared incompatible with BFO’s upper-level distinction into continuants and occurrents. The Clinical finding class seemed to be an umbrella for all kinds of entities of clinical interest, such as material entities, processes, states, dispositions, and qualities. This suggests the conclusion that Clinical finding would not be a suitable upper-level class from an BFO perspective. On closer inspection of the taxonomic links within this hierarchy and the implicit meaning derived thereof, it became clear that Clinical finding classes do not characterize the entity (e.g. a fracture, allergy, tumor, pain, hemorrhage, seizure, fever) in a literal sense but rather the condition of a patient having that fracture, allergy, pain etc. This gives sense to the current characteristic of the Clinical Finding hierarchy, in which complex classes are modeled as subclasses of their constituents. Most of these taxonomic links are inferred, as the consequence of the ‘role group’ design pattern, which is ubiquitous in SNOMED CT and has often been subject of controversy regarding its semantics. Conclusion: Our analyses resulted in the proposal of (i) equating SNOMED CT’s ‘role group’ property with the reflexive and transitive BFO relation ‘has occurrent part’; and (ii) reinterpreting Clinical Findings as Clinical Occurrents, i.e. temporally extended entities in an organism, having one or more occurrents as temporal parts that occur in continuants. This re-interpretation was corroborated by a manual analysis of classes under Clinical Finding, as well as the identification of similar modeling patterns in other ontologies. As a result, SNOMED CT does not require any content redesign to establish compatibility with BFO, apart from this re-interpretation, and a suggested re-labeling. Regarding the feasibility of harmonizing terminologies with principled foundational ontologies post-hoc, our results provide support to the assumption that this does not necessarily require major redesign efforts, but rather a careful analysis of the implicit assumptions of terminology curators and users. According to the Thomistic tradition, the Principle of Totality (TPoT) articulates a secondary principle of natural law which guides the exercise of human ownership or dominium over creation. In its general signification, TPoT is a principle of distributive justice determining the right ordering of wholes to their parts. In the medical field it is traditionally understood as entailing an absolute prohibition of bodily mutilation as irrational and immoral, and an imperfect obligation to use the parts of one’s body for the (...) perfection of the bodily whole. TPoT is thus a key element of the system of principles within which an individual exercises her right to life: it helps specify the nature, scope and limits of those actions by which an agent permissibly acts in order to preserve her life. While the Thomistic tradition and the Catholic Church have drawn clear conclusions from the principle regarding, e.g., direct sterilization and non-therapeutic experimentation on human subjects, less attention has been given to the implications of TPoT for non-therapeutic procedures that may positively impact biological functioning or supra-biological goals–that is, for human “enhancement.” We will explore the degree to which TPoT non-univocally guides our use of both artifacts and bodies. We will argue that a careful analysis of these distinct kinds of totalities suggests that the application of TPoT to artifacts and bodies is strongly isomorphic, which is what tempts advocates of the Principle of Autonomy to invalidly infer the absolute dominium of the individual over her body. The inference is invalid because this isomorphism also includes a principle of intrinsic value whose function is to resist the instrumentalization of both artifacts and bodies in some contexts; we are not even related to artifacts as advocates of absolute autonomy believe we are, let alone to our bodies. Rather, the limits of human dominium are determined by the nature and finalities, inherent or acquired, of the objects in question, and it will be argued that articulating these limits raises important, understudied, and fascinating questions about the permissibility of various kinds of human enhancement. G protein‐coupled receptors (GPCRs) constitute the largest family of transmembrane proteins and play a crucial role in regulating diverse cellular functions. They transmit their signaling via binding to intracellular signal transducers and effectors, such as G proteins, GPCR kinases, and β‐arrestins. To influence specific GPCR signaling behaviors, β‐arrestins recruit effectors to form larger signaling complexes. Intriguingly, they facilitate divergent functions for the binding to different receptors. Recent studies relying on advanced structural approaches, novel biosensors and interactome analyses bring us closer (...) to understanding how this specificity is achieved. In this article, we share our hypothesis of how active GPCRs induce specific conformational rearrangements within β‐arrestins to reveal distinct binding interfaces, enabling the recruitment of a subset of effectors to foster specialized signaling complexes. Furthermore, we discuss methods of how to comprehensively assess β‐arrestin conformational states and present the current state of research regarding the functionality of these multifaceted scaffolding proteins. Farming of fish and other aquatic species has increased in recent decades and never before have there been more controversial debates on animal welfare in fish husbandry. The practices used and associated welfare issues are becoming increasingly focused on by scientists, consumers and policy makers. International and national organisations have issued recommendations and guidelines concerning fish welfare but there is still a lot of information lacking. Due to § 2 of the German animal protection law, animals must be adequately nourished, (...) cared for and behaved in a proper manner, and pain or avoidable suffering or damage must be avoided. Keeping fish in accordance with the animal protection laws should be part of a good professional aquaculture practice and is indispensable for ethical, legal and economic reasons. As other vertebrates fish have the right of a healthy life with physical and mental integrity, under which they can perform adequately.The paper provides and discusses legal aspects of animal protection applied to fish taking the German welfare act and EU regulations as example. First possible indicators for the assessment of well-being will be mentioned. The assessment of fish welfare indicators should be carried out within the frame of daily and regular working routines, e. g. feeding, harvesting, grading or slaughtering. Further customer’s perception and human interpretation of welfare in fish are discussed and set into the context of human sentiments, projected on fish and the debate on consciousness in fish. Problem: Visions of technology, future scenarios, guiding visions represent imaginations of future states of affairs that play a functional role in processes of technological research, development and innovation—e.g. as a means to create attention, communication, coordination, or for the strategic exertion of influence. Since a couple of years there is a growing attention for such imaginations of futures in politics, the economy, research and the civil society. This trend concerns technology assessment as an observer of these processes and a consultant (...) on the implications of technology and innovation. TA faces increasing demands to assess imaginations of futures that circulate in the present and to participate in shaping these through scenarios or foresights. More than ever, this raises the question, which propositions can be made based on these imaginations by TA and how this can be used in advisory practices. Imaginations of futures are relevant for TA not as predictions but in their significance and effectiveness in the present, which need to be understood and assessed.Contents: This discussion paper outlines how present significance and effects of imagined futures in technological research and innovation processes can be conceived and analyzed. In this paper, all forms of imaginations of technology futures will be called “socio-technical futures” because within them technological developments and social changes are interwoven and inseparably interrelated. In this paper, we discuss why TA should analyze socio-technical futures, how such analyses can grasp the societal conditions that are expressed in the imagined futures and how these become effective in processes of technology development, communication, decision making etc. We raise the question which self-reflexive positioning or possible realignment of TA is needed as a response to its increased concern with assessing and even co-producing socio-technical futures. The latter is often demanded regarding the growing attention by politics and publics to imaginations of futures with wide temporal and spatial reach.Addressee of this paper is the TA community in a broader sense. The aim is to sensitize colleagues for the topic and its challenges, to consolidate discussions and to provide theoretical and methodical suggestions for research in TA and related advisory practices with respect to socio-technical futures. This paper has been originally initiated during the workshop “The present of technological futures-theoretical and methodical challenges for Technology Assessment”, in which all of the paper’s authors participated. In this chapter, Robb Dunphy is concerned with the nature of G.E. Schulze's scepticism as he presents it in his 1792 work Aenesidemus, and with its relation to the metaphysical projects of Kant, Reinhold, and later German Idealists. After introducing Schulze's text, Dunphy turns to a recent interpretation offered by Jessica Berry, who claims that the extent to which Schulze endorsed a genuinely Pyrrhonian Scepticism has gone unacknowledged, both by his idealist contemporaries and by the majority of the secondary literature (...) on the period. Berry suggests that this unacknowledged Pyrrhonism in Aenesidemus provides the resources for a more radical criticism of the German Idealists' scientific, systematic metaphysical ambitions, to which they remain fundamentally vulnerable. Despite agreeing that an exploration of Schulze's debt to Pyrrhonism represents a valuable addition to our understanding of his scepticism, in the latter parts of the chapter Dunphy suggests that the Berry's attribution of a Pyrrhonian Scepticism to Schulze should to some extent be rejected, and that such a scepticism is perhaps not quite as problematic for the projects of Kant and the German Idealists as she suggests. Daniel Breazeale, one of the greatest Fichte specialists, offers here four essential texts. First, the heart of this edition and of Fichte's philosophy, is Foundation of the Entire Wissenschaftslehre (literally "Doctrine of Science") of 1794–95. This central text has already been translated into English by Peter Heath and John Lachs under the title Science of Knowledge (New York: Appleton Century Crofts, 1970), but as Breazeale explains in his preface, he corrected the errors in translation and above all wished to propose a version that was "as broadly accessible as possible" (xi). This is a great challenge, given that this version of the Wissenschaftslehre (henceforth WL) is considered one of the most abstract and difficult texts in philosophy. Let us recall that the term 'WL' qualifies Fichte's entire undertaking and not just one of his works; Fichte thus professed more than fifteen versions of the WL.With this fundamental text, we are also offered a revised translation of two essays (which Breazeale had already edited in 1988): Concerning the Concept of the WL, or of So-Called 'Philosophy' (April–May 1794) and Outline of What is Distinctive of the WL with Regard to the Theoretical Power (winter 1794–95). Finally, Breazeale translates for the first time into English The Zurich WL, the lectures Fichte gave before a small committee from February 24 to April 26 of 1794. So, what question or problem was the source of his philosophy that he felt the need to rename WL? Fichte's initial problem was that of the foundation of transcendental idealism. The criticisms of skeptics like Gottlob E. Schulze (also known as "Aenesidemus") or Salomon Maimon had revealed the inadequacy of the foundations laid down by both Kant and Reinhold. As Fichte says and Breazeale reminds us, "The system has to be rebuilt" (4). This rebuilding is the common object of these four translated texts. Part One of Fichte’s 1794/95 Foundation of the Entire Wissenschaftslehre sets forth three basic principles (Grundsätze) as the founding claims of a ‘theory of science’ that should continue and consolidate Kant’s work by vindicating and integrating the theoretical and practical essentials of the Critical philosophy. These principles (my translations) are: (1) “The I originally absolutely posits its own being.” (2) “A not-I is absolutely opposed to the I”; ergo, “opposition in general is absolutely posited by the I.” (3) “In the (...) I, the I opposes a divisible not-I to the divisible I.” This essay explores the ways in which these principles both (i) structure Fichte’s own post-Kantian position and (ii) seek to neutralize some important criticisms of the broader Kantian project. In particular, I argue that Fichte’s principles draw upon a distinctively Kantian conception of pure-rational activity, as the autonomous origination and instatement of pure order-inducing forms: non-sensory notions that inform all truth-apt cognition, and affect-independent norms that orient all autonomous volition. Accordingly, the principle, “the I originally absolutely posits its own being” describes the transcendentally most basic instance of such activity: that pure-rational act upon which all other such acts – ergo, all articulate cognition and all principled agency – ultimately depend. For Fichte, this is an act in which the I conceptually positions rational activity as such, in its strict purity and unmitigated autonomy, as both essentially constitutive of and unconditionally normative for its own existence. Thus the first principle of Fichte’s system figures the most basic pure-rational act as both theoretically determinative and practically legislative – with the consequence that even our most basic cognitive accomplishments depend upon a broadly ethical vocation intrinsic to rationality itself. After making the case for that interpretation, I also, more briefly, discuss the ways in which Fichte’s principles underwrite his responses to some trenchant criticisms of the broader Kantian project: the ‘Humean skepticism’ of G. In Aenesidemus, G.E. Schulze adopts the skeptical voice of Aenesidemus and engages in critical dialogue with Hermias, a Kantian, in the hopes of laying bare what he views as the fundamental issues of K.L. Reinhold’s version of critical philosophy. While some attacks reveal a deep misunderstanding of Reinhold’s Elementarphilosophie on Schulze’s part, others hit their mark. In the Aenesidemus Review (1794), J.G. Fichte at times agrees with criticisms raised by Aenesidemus and at times defends Reinhold against them. On Fichte’s view, (...) Schulze succeeds in proving that the first principle of Reinhold’s Elementarphilosophie, the principle of consciousness (Satz des Bewußtseins), is neither self-evident nor self-determining. Therefore, it cannot be the first principle of philosophy. However, Schulze fails to dissuade Fichte from viewing Reinhold’s principle of consciousness as the pithiest expression of human consciousness of the time. For these reasons, Fichte holds that Reinhold’s principle of consciousness must be deduced from an even higher principle. The study of the “first wave” of reactions to the Critique of Pure Reason in Germany from the second half of the 1780s until the beginning of the nineteenth century reveals the paradoxical status of the Kantian transcendental subject. While the existence of the transcendental subject, whatever the term means, is not open to question since it arises from the very essence of critical philosophy, the fundamental status of the subject is sometimes questioned in this period. Although the meaning of (...) the concept of transcendental subject seems obvious today it lends itself to various interpretations in the late eighteenth century. To achieve my goal I have undertaken a textological analysis of the works of the earliest opponents and followers of the Kantian critique and a reconstruction of the conceptual field in the midst of which the transcendental subject has been planted. Among others I draw on the works of J. S. Beck, J. A. Eberhard, J. G. Hamann, F. H. Jacobi, S. Maimon, K. L. Reinhold, G. E. Schulze and A. Weishaupt. The authors of the period are grouped depending on the common themes and questions that prompted them to turn to the concept of the transcendental subject, even though the results of their reflections did not always coincide. These authors think of the transcendental subject in its relationship to the transcendental object, or as “something = х”, and in terms of the relationship of representation to the object. It is characterised sometimes as something absolutely hollow, and sometimes as the fullness of true reality. The status ascribed to the transcendental subject is sometimes that of a thing-in-itself and sometimes that of a “mere” idea. Finally, Kant’s transcendental subject was sometimes seen as something to be overcome and sometimes as an infinite challenge to understanding. (shrink)

G. Fichte has of late displayed a rather substantial, and even remarkable, transformation. Where before Fichte was viewed—and most often dismissed—as advancing an unpalatable type of metaphysical idealism, in recent years several new perspectives on Fichte have emerged, each claiming to improve on (...) the old, standard picture. 1 The transformation of Fichte's image has been accompanied—or, more precisely, has been made possible—by a renewed interest in the relatively forgotten figures in the interregnum between Kant and Hegel. Fichte's thought is now understood in the context not only of Kant's transcendental idealism, but of, among others, Reinhold's Elementarphilosophie, Aenesidemus-Schulze's skepticism, Jacobi's intuitionism, and Beck's Standpunktslehre. By casting Fichte in this light, a more nuanced understanding of his project has been developed, one that does not simply follow the [End Page 101] traditional view of the Wissenschaftslehre as a full-blown metaphysical idealism in which the world of objects is understood as the product of an Absolute I.But while this 'contextual' approach has led to a renewed appreciation of the Wissenschaftslehre, I will suggest that the new focus on Fichte's historical milieu is incomplete. A lacuna in the current understanding of Fichte can be traced to a neglect of Salomon Maimon, an idiosyncratic and rather obscure critic of Kant. In this paper, I will argue that taking Maimon's skepticism into account when interpreting Fichte's idealism requires a rethinking of Fichte's project. In particular, in order to respond to Maimon's skepticism—an avowed project of the Wissenschaftslehre—Fichte's position must advance an idealist metaphysics, one in which the content of cognition is taken to be constituted by an act of the absolute I. This claim, of course, might seem rash, since it threatens to saddle Fichte with a gratuitous form of absolute idealism. It thus seems to run counter to recent attempts to make Fichte into a figure appealing to modern eyes. Such a concern, however, is I think misplaced. Instead, I will argue that Fichte's idealism, although perhaps ultimately unsuccessful, nevertheless should be of great interest precisely because it stands as an important attempt to provide an answer to a question—the nature of 'givenness'—that remains central to epistemology. 2The paper is divided into five parts. The first presents the historical evidence for Maimon's influence on Fichte. Part 2 offers a discussion of what I call 'Maimon's challenge' to the Kantian notion of 'discursive' cognition, as well as the ways in which this served as a goad to the development of the Wissenschaftslehre. In Part 3 I turn to a close reading of §1 of the Grundlage of 1794, where I try to show how a recognition of Maimon's challenge can help make sense of the foundations of the Wissenschaftslehre's project, and in particular the 'category of reality,' an otherwise highly obscure step in Fichte's argument. Part 4 offers a short discussion of the ways in which a Maimonian-minded account of §1 contributes to the understanding of the Grundlage as a whole. The notion that there exists a fundamental need to exert control over or to influence one’s environment has enjoyed a long history in psychology (e.g., DeCharms, 1968; Heider, 1958) and has stimulated considerable theoretical work. Such a need has been characterized by theorists at multiple levels of analysis. Control motivation, for example, has been characterized broadly in terms of proactive (White, 1959) or reactive (e.g., Abramson, Seligman, & Teasdale, 1978; Brehm, 1966; Brehm & Brehm, 1981) strivings for control over general (...) or specific (Brehm & Brehm, 1981) and central or peripheral outcomes (Thompson, 1993). Additionally, various types of control strategies used to gain or maintain a sense of personal control have been proposed (e.g., Averill, 1973; Heckhausen & Schulz, 1995; Rothbaum, Weisz, & Snyder, 1982; Thompson, 1981). Modes of control, for instance, have been categorized as either primary or secondary. Primary strategies involve direct action undertaken to produce desirable and avoid undesirable outcomes in the external world, whereas secondary strategies employ primarily cognitive processes undertaken to produce a change within the person. Recently, Heckhausen and Schulz (1995) have further delineated these primary and secondary forms of control according to whether they are based on veridical or illusory causal understandings of the world and whether they are functional or dysfunctional. While most control theorists view primary control as preferable to secondary control, the latter is viewed as critical in the process of adaptation to control failures and in the promotion of future primary control attempts.
